FLT3 (FMS-like tyrosine kinase 3) is a receptor tyrosine kinase preferentially expressed on leukemic cells of the myeloid lineage. It belongs to class III tyrosine kinase family and is expressed in some normal hematopoietic cell types and plays an important role in the pathogenesis of AML (Acute Myeloid Leukemia). FLT3 mutations can be divided into 2 categories: internal tandem duplications (FLT3/ITD mutations) in or near the juxtamembrane domain of the receptor and point mutations resulting in single amino acid substitutions occurring within the activation loop of the tyrosine kinase domain (FLT3/TKD mutations). An FLT3/ITD mutation varies according to age and clinical risk group, being less common in pediatric AML and in AML arising from an antecedent myelodys-plastic syndrome.
