It is also recognized as a significant stress response gene and is regulated by a number of stress stimuli. These include iron chelation, nickel compounds, cobalt, hypoxia, oxidative stress, and tunicamycin. NDRG1 has been shown to inhibit metastasis and tumor progression via its effects on a variety of critical cell signaling pathways involved in proliferation, locomotion, and oncogenesis e.g. AKT, TGF-β, ROCK1/pMLC2, Ras, WNT, etc. NDRG1 is predominantly cytoplasmic, and the protein is ubiquitously expressed both in normal and neoplastic tissues. Additionally, it is highly conserved among multicellular organisms. NDRG1 missense mutations causes HMSNL (Hereditary Motor And Sensory Neuropathy-Lom), an autosomal recessive form of CMT (Charcot-Marie-Tooth disease) also called CMT4D. Further, NDRG1 has been shown to have a role in myelin sheath maintenance exocytosis in mast cells differentiation and NDRG1 expression is induced by homocysteine, androgen, as well as in response to various forms of cellular stress such as hypoxia , heavy metals or DNA damage.
