SLC34A1, a type IIa sodium-dependent phosphate cotransporter, is a multi-trans membrane protein which plays a critical role in the rate-limiting step of renal Pi reabsorption. The Pi transport activity in the proximal tubule is dependent on the amount SLC34A1in the apical plasma membrane. SLC34A1 ablation leads not only to hypophosphatemia but also to hypercalciuria and high levels of 1,25-dihydroxyvitamin D3 [1,25(OH)2D3] and skeletal abnormalities. It has been reported that renal SLC34A1 gene ablation induces renal calcification due to abnormalities of mineral metabolism. Renal SLC34A1 expression is regulated by dietary Pi and hormones such as 1,25(OH)2D3, PTH (parathyroid hormone), FGF-23 (fibroblast growth factor- 23), dopamine, thyroid hormones, and glucocorticoids.
