Tyrosine 3-monooxygenase is a non-heme Fe enzyme found in the brain and adrenal gland of humans that catalyzes the hydroxylation of the amino acid L-tyrosine to form L-DOPA (L-3, 4 -Dihydroxyphenylalanine). This reaction is the rate-limiting step in the biosynthesis of the catecholamine neurotransmitters dopamine, epinephrine and norepinephrin, making it vital to nervous system function. This protein is located, on chromosome locus 11p15, contains 13 exons/12 introns spanning approximately 8 kbp. Although a single-copy gene, it produces four different types of mRNA through alternative splicing of a single primary transcript indicating one novel level of regulating catecholamines. Substantial loss of tyrosine 3-monooxygenase enzymatic activity has profound consequences both in humans and in mice with targeted ablation of the tyrosine 3-monooxygenase locus (Ref. 3). Missense mutation in both alleles of the tyrosine 3-monooxygenase gene is known to cause dopamine-related phenotypes, including dystonia and infantile Parkinsonism.
