IL-23, a member of the IL-12 cytokine family, is composed of a p40 subunit, which is shared with IL-12, and a p19 subunit, with an approximate molecular weight of 60 kDa. IL-23 is produced by activated dendritic cells and macrophages in response to microorganisms. IL-23 promotes Th17 cells producing TNF-a, IL-17, IL-6, IL-22, GM-CSF, and other novel factors, which are associated with the induction of autoimmune inflammation. IL-23 is essential for Th17 differentiation, expansion, and survival by binding to its receptor, thereby activating the signaling pathway. Many studies revealed that the IL-23/Th17 pathway is implicated in the pathophysiology of various autoimmune diseases, such as autoimmune arthritis, primary biliary cirrhosis, and inflammatory bowel disease.
