NY-ESO-1, a member of the CT (cancer/testis) family, is a tumor-specific shared antigen with distinctive immunogenicity. NY-ESO-1 is expressed in testis and ovary and in a wide variety of cancers. It is also detected in uterine myometrium. The expression is from 18 weeks until birth in human fetal testis. Furthermore, in the adult testis, NY-ESO-1 is strongly expressed in spermatogonia and in primary spermatocytes, but not in post-meiotic cells or in testicular somatic cells (at protein level).The molecular basis of T cell-mediated antitumor immunity has been elucidated by the identification of a number of tumor antigens recognized by CD8+ T cells. Among these antigens, NY-ESO-1 is of particular interest because both the cytotoxic T lymphocyte and antibody have been shown to react with this antigen.
